Jul 28, 2020 · AMP-activated protein kinase (AMPK) phosphorylation of ACE2 enhances ACE2 stability. This mode of posttranslational modification of ACE2 in vascular endothelial cells is causative of a pulmonary hypertension (PH)–protective phenotype.
AMP-Activated Protein Kinase Induces p53 by Phosphorylating …
AMP-activated protein kinase (AMPK) has been shown to activate p53 in response to metabolic stress. However, the underlying mechanisms remain unclear. Here we show that metabolic stresses induce AMPK-mediated phosphorylation of human MDMX on Ser342 in vitro and in cells, leading to enhanced association between MDMX and 14-3-3.
AMPK reverses the mesenchymal phenotype of cancer cells by ... - PubMed
Sep 1, 2014 · Here, we report a novel function of the master metabolic kinase AMPK in suppressing EMT by modulating the Akt-MDM2-Foxo3 signaling axis. This mechanistic link was supported by the effects of siRNA-mediated knockdown and pharmacologic activation of AMPK on epithelial and mesenchymal markers in established breast and prostate cancer cells.
AMP-activated protein kinase induces p53 by phosphorylating ... - PubMed
Here we show that metabolic stresses induce AMPK-mediated phosphorylation of human MDMX on Ser342 in vitro and in cells, leading to enhanced association between MDMX and 14-3-3. This markedly inhibits p53 ubiquitylation and significantly stabilizes and activates p53.
MDM2 Inhibitor, Nutlin 3a, Induces p53 Dependent Autophagy in …
We investigated autophagy induction in acute leukemia by Nutlin 3a, a first-in-class MDM2 inhibitor. Nutlin 3a induced autophagy in a p53 dependent manner and transcriptional activation of AMP kinase (AMPK) is critical, as this effect is abrogated in AMPK -/- …

Degradation of AMPK by a Cancer-Specific Ubiquitin Ligase
Oct 7, 2021 · Here, we describe a widespread mechanism to suppress AMPK through its ubiquitination and degradation by the cancer-specific MAGE-A3/6-TRIM28 ubiquitin ligase. MAGE-A3 and MAGE-A6 are highly similar proteins normally expressed only in the male germline, but frequently re-activated in human cancers.

Ketogenic diet induces p53-dependent cellular senescence in …
May 17, 2024 · Our experiments have shown that a KD can induce p53 signaling through AMPK activation combined with inactivation of MDM2 by caspase-2 cleavage, ultimately leading to an increase in p21 and cellular senescence in multiple organs (fig. S9).
Degradation of AMPK by a Cancer-Specific Ubiquitin Ligase
Feb 12, 2015 · A cancer-specific E3 ubiquitin ligase ubiquitinates and degrades AMPK, resulting in downregulation of autophagy and increased mTOR signaling. This regulatory axis demonstrates how altered cellular metabolism can act as an oncogenic driver in cancer.
